Project Summary/Abstract The new concept of a transient focal adhesion (FA) that forms and then rapidly disassembles has emerged as a new paradigm in cell biology. This new concept is particularly relevant for the better understanding of processes such as cell migration and invasion where cells physically attach to their substratum only transiently. This concept is supported by extensive evidence showing that the transient adhesion of cells to the ECM is associated with the transient activation of integrins and their downstream effector proteins. Based on this evidence and on the importance of integrins in cancer progression, the concept of the transient focal adhesion is extended to investigate the involvement of the transient adhesome in cancer metastasis. In this model, we hypothesize that the process of metastasis is initiated by the transitory interaction of tumor cells with ECM proteins that triggers integrin activation. The focal adhesion of cancer cells transiently interacts with ECM proteins to form a favorable microenvironment for cancer cells that can develop subsequent steps of metastasis. In this project, we intend to identify protein microenvironment for the FA of cancer cells during cancer progression and investigate their roles in cancer metastasis. In Aim 1, we will identify ECM proteins that interact with the transient FA of cancer cells during cancer progression and metastasis. We will use super-resolution imaging and fluorescence resonance energy transfer (FRET) approaches to visualize the protein microenvironment of the transient FA in live cells. We will also use multiplexed mass spectrometry to identify candidate proteins that interact with the transient FA of cancer cells. We will validate the interaction between the transient FA and candidate ECM proteins using super-resolution imaging and live cell-based FRET assays.
